Students are required to attend all sessions in the practical course. The final grade will be evaluated from the gradings of class participation, including pre-class learning and laboratory notes, and the reports.

概要

The aim of this practical course is to understand the function of genes, the blueprint of life. Students observe that a phenotype is determined by a genotype, and work on DNA to learn its molecular characters. Students also learn the techniques of molecular biology from principles, which is essential for modern life sciences. Experiments include microbiology, extraction, purification and analysis of DNA, molecular cloning, gene expression, and protein crystallization. Please note that the course has three parts, and the order among them may change from the schedule. This laboratory course is one of Department Core Courses and Elective compulsory courses in Green Science course (for third-year student or higher). Students will acquire the ability to contribute to the creation of functional biomolecules, and technology development in related fields (Curriculum Policy #3).

目標

Students experience and understand biological activities at molecular level, and obtain the basic techniques of molecular biology. The aims of this laboratory course are to; 1) Learn safety and ethics in biological research 2) Acquire the ability to understand phenomena of life based on the fundamentals of biology. 3) Acquire the ability to systematically understand the fundamental of materials and life, and contribute to the creation of functional biomolecules as well as technology development. (Diploma Policies #2, 3).

授業外の学習

Students are required to complete pre-lab learning before each session (1 hour) and revise experimental procedures and the obtained results after each session (1 hour). For each part a laboratory report needs to be submitted.

  1. Introduction and safety guidance (Class 1)
  2. PART-1: Basic procedures (Classes 2 to 6) Aseptic procedure, Media preparation, Culturing bacteria, Plasmid DNA extraction, Agarose gel electrophoresis, and Restriction enzyme digestion of the plasmid DNA
  3. PART-2: Molecular cloning of green fluorescent protein (GFP) gene (Classes 6 to 10) PCR amplification and purification of the GFP gene, cloning, transformation of bacterial cells, observation and culturing of GFP-positive bacterial colonies and purification of GFP
  4. PART-3: Crystallisation of protein (Classes 11 to 14) Crystallization of lysozyme protein, and observation and evaluation of the crystals.
